Draper Nanotechnology Could Fight Influenza, Other Viruses

Newly emerging flu viruses could soon be countered by a treatment that Draper Laboratory is developing that traps viruses before they can infect host cells.


Further into the future, patients suffering from any type of virus could be cured with DRACO, a drug also under development at Draper that is designed to rapidly recognize and eliminate cells infected by virtually any virus.


Both methods could help safeguard against bioterrorist attacks and naturally occurring pandemics in a manner that is unlikely to lead to treatment-resistant strains. Initial testing on the treatments, which each use tiny, non-toxic particles that can be injected, inhaled, or eaten, has shown them to be effective and safe against a multitude of strains of disease.


Nanotraps, which could be taken at the first sign of infection or exposure, is likely the first of the products ready for use, and is expected to begin clinical trials in two to five years, according to Jim Comolli, who leads the research on the effort at Draper.


Nanotraps, developed by a team of researchers from Draper, MIT, the University of Massachusetts Medical School, and the University of Santa Barbara, are nanoparticles that act as viral traps using specific molecules found naturally within the human body. A paper published in the March issue of Journal of Biological Chemistry detailed their findings.


The nanotraps look like the surface of a cell, with numerous carbohydrate molecules attached that closely resemble those targeted by flu viruses in the human respiratory system. These molecules, initially characterized in the Sasisekharan Lab at MIT, act as bait for the flu virus, which bind to the nanotrap instead of a host cell and are cleared away with mucus, preventing infection, Comolli said.


The research team has demonstrated in the laboratory that the nanotraps effectively countered multiple influenza strains able to infect humans and went on to show nanotraps protected mice infected with the flu. They have also developed additional particles geared toward other types of respiratory viruses.


Nanotraps, unlike most vaccines, are not strain specific and are designed to be effective against newly emerging strains of human-adapted influenza virus. Since nanotraps mimic a fundamental step in the viral life cycle the binding of the virus to a host cells receptor nanotraps may offer an opportunity to treat devastating infectious diseases without causing the development of treatment resistance, Comolli said.


The nanotrap molecule is composed mainly of compounds found naturally in the human body so it is likely to be safe as an inhalant, topical solution, or intravenous treatment and inexpensive to manufacture. With further development, nanotraps have the potential to treat a large range of infectious diseases because the attached carbohydrates can be tuned for other viruses such as HIV, Respiratory Syncytial Virus (RSV), and Herpes Simplex Virus (HSV), as well as bacteria and toxins.


The work was originally funded by the Defense Advanced Research Projects Agency (DARPA) to address emerging pandemics or bioweapons targeting US troops; Draper is continuing the research with the intent of applying it more broadly to the civilian community.


While nanotraps could be customized to treat a variety of viruses, another project underway at Draper, DRACO could do so with a single approach. DRACO is a unique therapeutic drug that should be effective against a very broad spectrum of viruses, just as existing antibiotics are effective against a broad range of bacterial infections. DRACO could be ready for human clinical trials in five to 10 years, according to Todd Rider, who leads the research at Draper.


DRACO (Double-stranded RNA Activated Caspase Oligomerizer) is designed to detect cells that have been virally infected and then eradicate only the infected cells, rapidly ending the infection. DRACO has proven effective in vivo against influenza and three hemorrhagic fever viruses, and in vitro against 15 different viruses including common cold viruses, the H1N1 influenza strain, adenoviruses, a mouse polio virus, dengue fever, and stomach viruses, among others. It has also been tested and proven safe in both mice and 11 different human and animal cell types representing organs like the heart, lungs, liver, and kidney, among others.


DRACO is designed to be attracted to a specific type of RNA exclusive to viral infections long double-stranded RNA, or dsRNA. Detecting this dsRNA in a human or animal cell indicates that that host cell has been taken over by a virus and is now in the process of creating more viruses. DRACO enters cells and attaches itself to any dsRNA. Once two or more DRACOs attach to the dsRNA, they interact with one another and activate a natural self-destruct switch inside the infected cell, terminating the infected cell and the virus that it was helping to reproduce.


DRACO is designed to be both broad-spectrum and nontoxic to humans overcoming existing issues with current anti-viral treatments. Because DRACO is so broad-spectrum and acts so rapidly, there is little opportunity for the virus to evolve defenses against this treatment, Rider said.


Rider is expanding his DRACO research and testing more strains of various viruses in cells and animals.


DRACO has been funded by grant AI057159 from the National Institute of Allergy and Infectious Diseases and the New England Regional Center of Excellence for Biodefense and Emerging Infectious Diseases, with other funding coming from DARPA, the Defense Threat Reduction Agency, and the office of the Director of Defense Research and Engineering.

When you play paintball, you would like to protect your face. And by wearing a paintball mask, you can do exactly just that.


A mask is an important protective gear worn when playing paintball. It protects your face from getting hit by a paintball marker that travels up to 300 feet per second. It would cost less than $ 20 to $ 100. It may also come with accessories like headbands and visors to shield you from the sun's glare.


A paintball mask basically protects your face from paintball markers. It also prevents paint from getting in your eyes, nose, ears or mouth. Paint in a paintball marker is toxic and must not be ingested. It also prevents paintball markers from hitting any sensitive part of your face like your nose or your eyes or any part that can be irritated by the paint.


A good mask also helps prevent head injuries when you fall or trip during the game. A paintball mask must have adequate padding that fits the head snugly into the mask. The padding must contour your face and your head when you wear it but return to its original shape when you take it off. It must also be firmly glues onto the plastic of the mask. You must be able to tighten or loosen the straps of the mask without stretching it too much. Your mouth guard must not hit your chest when you look down.


A good paintball mask should also have a thermal or anti-fog lens. This is composed of two lenses with a small air pocket in between that stabilizes the heat on from your face and outside of the mask, preventing fogging on the lens. You could also opt to install fans on your mask, though not all masks readily allow fans to be installed into them. These fans go on top of the lens and help moisture from your face evaporate.

These fans are pretty expensive and would require some batteries for it to operate. It also makes considerable noise and is not very durable.

Keeping your mask clean is a must. First, you can run a rag over the mask to take out most of the paint on your mask. You can spray water on the paint to loosen it a little. Do not use any chemical or lens cleaner on your paintball mask. This may be harmful to your lens and could ruin it.


Make sure that you clean the creases and little holes in your paintball mask. It stops the paint from building up in that space and trickling down on your face or your lens later on. The lens on your mask is detachable. Remove it and clean the edges of your lens. Wipe the lens with a microfiber cloth to absorb all the excess oil on the lens of your paintball mask. This will give you clear vision when you use it next time.

Intrapreneurs, like their entrepreneurial counterparts, need to envision and create an idea. But instead of convincing outside investors and risking their own personal capital, the intrapreneur needs to create the right team within his company to successfully launch the new products. Then the intrapreneur needs to convince some "mover and shaker" in senior management to approve the new product concept and then to invest the company's time, money, and other resources.


Some enlightened corporations, such as 3M, do allow an employee-intrapreneur limited freedom to pursue new ideas. The Intrapreneur still has too overcome the roadblocks of formal corporate structure and the delays of corporate bureaucracy. The corporate entrepreneur, or Intrapreneur, must convince senior management that the new, out of the main stream idea has merit, has possible market, and that the new produce would be both profitable and synergistic to the corporation.


The good new is that corporate entrepreneur or Intrapreneur is really just an entrepreneur who has less risk in building a new venture. The Intrapreneur has materially less control of when, or even whether, the product will be launched by his employing corporation. To be a successful Intrapreneur takes a unique set of talents and abilities far more than just being creative. The must be willing to take some personal risks at sharing and pushing an unique idea with the senior management of the corporation. The Intrapreneur must have the persistence and perseverance to work to secure senior management's approval to create and launch the product or service.

The successful Intrapreneur must have the personal drive to see the project to a successful fruition, no matter what it may take.

Intrapreneuring and intrapreneurship existed in corporations for decades, even before the term was used in the 1982's. Gifford Pinchot (1985) made it famous in his book Intrapreneuring. Apple's Chairman, Stephen Jobs (1985) use the term "intrapreneurship" in his article Newsweek article in the September of 1985. Job's said, "The Macintosh team was what is commonly known as intrapreneurship-only a few years before the term was coined - a group of people going, in essence, back to the garage, but in a large company." Back in 1982, Dr. Haller wrote his master thesis on the "Intrapreneurship Success" of PR1ME Computer Inc in the late 1970's and early 1980's.


Some major public companies specifically foster and encourage intrapreneurial explorations on company time through formal corporate intrapreneurship programs. During the intrapreneurial journey from idea to product, some ideas and products are ignored or neglected. The twelve year course from idea creation to production of product for 3M'sPost-it Notes™ is an example of this.


Just like the entrepreneur, the intrapreneur must constantly stay focused and fully committed to the product and be willing to keep pushing, even if it takes years to produce their new product and deliver it to market!

Through it's intrapreneurial "Bootlegging Policy," (which allows employees to spend up to 15 percent of their time at work developing their own creative ideas for the betterment of the company and the creation of new products or services) the usually staid, 3M Corporation was able to capitalize on the creative ideas of 3M Intrapreneurs Spencer Silver and Art Fry to create and launch the 3M Post-it Note product. 3M scientist Spencer Silver invented a not so sticky adhesive in 1968, but it took him several years to come up with how to best use and market the product. Spencer shared his idea through seminars in which he shared the features (and benefits) of this semi-adhesive to fellow 3M employees.


But no one really caught the vision of his product for five long years. Finally, a co-worker, Art Frey, recognized the need for Spencer's idea! During choir practice and performances, Frey's bookmarks were regularly falling out of his hymn book. This product concept solved his problem! Still, 3M still did not do anything with Spencer's idea for over a decade. Then finally, in 1980, Minnisota Mining and Manufacturing's (3M) Post-its Notes product line, which is their moveable semi-sticky notes product, was successfully launched across the entire US, and internationally.

The Debate Surrounding Diesel Particulate Filters


There have been enormous technological advances ever since the industrial revolution and these have affected and influenced almost every aspect of society. The transport sector in particular has seen enormous changes over the years, and technological progress with regard to motor vehicles in particular has had major impacts. In today's world, whilst concepts such as 'value' matter immensely to consumers, the real keystones are emissions and economy.


This has had some dramatic effects and the whole pattern of vehicle usage has changed quite dramatically. One of the major areas in this regard is diesel engines. Once the preserve of heavy goods vehicles and big coaches, the diesel engine has found a place in normal passenger cars due to its combination of fuel economy, potential performance, and reliability. The proportion of diesel-powered cars on the roads throughout Europe in particular has grown beyond anyone's expectations over the last twenty or thirty years and diesel vehicles now account for some 30 per cent of sales.


The diesel engine offers many benefits, the biggest of which is its ability to produce comparatively high power outputs for relatively low fuel consumption. As a result, overall exhaust gas emissions are favourably low. However, there is one particular issue with diesel emissions, which is particulates. Diesel fuel tends to burn incompletely and produce carbon in relatively large quantities. This is discharged in the engine exhaust system in the form of soot particles, which are not only unpleasant but are known to have potential health issues.


Laws passed in various countries worldwide are aimed at reducing emissions from vehicles, and one aspect of these is to target particulates.

Accordingly, manufacturers have looked into various methods and most have added a Diesel Particulate Filter (or DPF) to car and truck exhaust systems. Put simply, a DPF is a basic mechanical filter that merely removes soot particles from the exhaust gas.Of course, there are downsides to this. The main issue is that the filter becomes loaded with carbon particles relatively quickly and unless there is a method of cleaning the filter, it soon becomes blocked to the extent of causing engine damage and massively affecting performance. For passenger vehicles, the method of 'regeneration' involves driving the vehicle at comparatively high speed for a period of time, which results in the filter becoming hot enough to burn off the accumulated deposits.

Anyone 'thinking outside the box' will quickly come to the conclusion that whilst there are obvious benefits to removing particulates from diesel exhaust gas, using a DPF has inherent disadvantages. Firstly, the use of a filter on its own reduces efficiency, as it forms a barrier to exhaust flow and therefore reduces efficiency and increases fuel consumption. Secondly, and more importantly, having to run the vehicle at higher speeds to burn off the deposited carbon is inherently wasteful and the end result is an increase I overall carbon dioxide emissions, which are themselves the target of worldwide reduction efforts.


The irony of the situation is that the vast majority of modern diesel engines can, with accurate electronic management systems, run at extremely high efficiency and massively reduced carbon emissions. A correctly managed diesel can easily meet the most stringent particulate emission targets without the need for a DPF at all, and with corresponding benefits to efficiency and therefore fuel consumption. So why don’t manufacturers do this? The answer lies in economies of scale. Car makers have to produce vehicles that are sold in many different countries, where the quality of fuel can differ dramatically. One management computer cannot deal with this, and the cost involved in programing individual vehicles for individual markets is significantly higher than buying DPF units in bulk. Production may also have to be diverted from one market to another depending on demand, causing even greater issues.


For the private motorist already running a diesel car, the situation is of course somewhat different. In the vast majority of cases, it is possible to simply remove the DPF unit intact. Carried out properly, regarding DPF removal can provide reduced fuel consumption and increased performance, with no downsides. However, the key word here is 'properly'. It is essential for the engine management computer to be comprehensively reprogrammed in accordance with local conditions. Anything else is a bodge that could actually do far more harm than good. However, properly undertaken, DPF removal may well be the best option for many drivers and owners.
